Purification and Properties of Chalcone Synthase from Cell Suspension Cultures of Soybean
نویسندگان
چکیده
Incubation of soybean cell suspension cultures in B-5 medium containing 0.4 m sucrose caused induction of chalcone synthase (CHS) up to a specific activity of 8 |ikat/kg. CHS from these cultures was purified to apparent homogeneity by a 5-step procedure. Isoelectric focussing of pure CHS gave a major band at pH 5.45 and two weaker bands at pH 5.35 and 5.5. At least the bands at pH 5.35 and 5.45 had CHS activity. Analysis of pure CHS by two-dimensional electrophoresis gave a set of six proteins with a Mr o f 40 kDa and pis between 6.0 and 6.6. One-dimensional PAG E of CHS under non-denaturing conditions gave three closely spaced protein bands. A specific antibody was raised against soybean CHS which cross-reacted with parsley CHS. Attem pts to find synthesis o f deoxychalcone or of the corresponding 7,4'-dihydroxyflavanone with CHS of different purification stages and with various cofactors failed.

Purification and properties of a stilbene synthase from induced cell suspension cultures of peanut.
Stilbene synthase ( resveratrol -forming) converts one molecule of rho- coumaroyl -CoA and three molecules of malonyl-CoA into 3,4',5- trihydroxystilbene . Following selective induction of stilbene synthesis in cell suspension cultures of peanut (Arachis hypogaea), the enzyme was extracted and purified to apparent homogeneity by chromatography on DEAE-cellulose and hydroxylapatite. The enzyme w...

Acridone synthase has been purified from cell suspension cultures of Ruta graveolens using a combination of gel filtration and ion exchange chromatography. The purified enzyme has an apparent molecular weight of 69 kDa on gel filtration and a subunit structure on SDS-PAGE of 40 kDa. The apparent Km-values are 10.64 microM and 32.8 microM for N-methylanthraniloyl-CoA and malonyl-CoA, respectivel...
متن کاملRapid Response of Suspension-cultured Parsley Cells to the Elicitor from Phytophthora megasperma var. sojae: INDUCTION OF THE ENZYMES OF GENERAL PHENYLPROPANOID METABOLISM.
Large and rapid increases in the activities of two enzymes of general phenylpropanoid metabolism, phenylalanine ammonia-lyase and 4-coumarate:CoA ligase, occurred in suspension-cultured parsley cells (Petroselinum hortense) treated with an elicitor preparation from Phytophthora megasperma var. sojae. Highest enzyme activities were obtained with an elicitor concentration similar to that required...
متن کاملEffects of fungal elicitor on lignin biosynthesis in cell suspension cultures of soybean.
Soybean (Glycine max L.) cells cultured in B5 medium produce extremely low amounts of lignin. However, modification in the growth medium, by lowering the concentration of NO(-) (3) and PO(2-) (4), results in the lignification of these cells without affecting levels of cell wall-esterified 4-coumaric and ferulic acid. The production of an extracellular, macromolecular complex by the cultured soy...
